Les \u00ab\u00a0mini-cerveaux\u00a0\u00bb qui en r\u00e9sultent sont devenus son syst\u00e8me mod\u00e8le pour explorer le comportement d&rsquo;une prot\u00e9ine cl\u00e9 dans le d\u00e9veloppement de la maladie de Parkinson, ce qui pourrait \u00e0 son tour ouvrir la voie \u00e0 des m\u00e9canismes susceptibles de stopper ou m\u00eame d&rsquo;inverser ce processus. <\/h4>\n\n<p class=\"wp-block-paragraph\">Afin d&rsquo;analyser les caract\u00e9ristiques biochimiques sp\u00e9cifiques du cerveau, la chercheuse postdoctorale Nguyen-Vi Mohamed de l&rsquo;Universit\u00e9 McGill utilise un mod\u00e8le constitu\u00e9 de cellules c\u00e9r\u00e9brales qu&rsquo;elle a cultiv\u00e9es dans son laboratoire. Ces \u00ab\u00a0mini-cerveaux\u00a0\u00bb, comme elle les appelle, reproduisent la structure du m\u00e9senc\u00e9phale humain et montrent les fonctions complexes responsables du fonctionnement de cet organe myst\u00e9rieux &#8211; ou, dans le cas d&rsquo;une maladie neurod\u00e9g\u00e9n\u00e9rative comme la maladie de Parkinson, de l&rsquo;arr\u00eat progressif de son fonctionnement. <\/p>\n\n<p class=\"wp-block-paragraph\">La recherche de Mohamed est rendue possible gr\u00e2ce \u00e0 une bourse de recherche fondamentale du Programme de recherche Parkinson Canada, et financ\u00e9e en partenariat avec le Fonds de recherche du Qu\u00e9bec &#8211; Sant\u00e9 (FRQS) pour un montant de 50 000 $ sur deux ans.<\/p>\n\n<p class=\"wp-block-paragraph\">\u00ab\u00a0Pour les cultiver, l&rsquo;id\u00e9e est d&rsquo;imiter ce qui se passe au cours du processus de d\u00e9veloppement\u00a0\u00bb, explique Mohamed, qui est all\u00e9 bien au-del\u00e0 d&rsquo;un simple r\u00e9seau plat de cellules dans une bo\u00eete. \u00ab\u00a0Il s&rsquo;agit d&rsquo;un mod\u00e8le 3D complexe avec diff\u00e9rents types de populations cellulaires. En 3D, les cellules ont des fonctionnalit\u00e9s plus matures, gr\u00e2ce \u00e0 la diversit\u00e9 des populations cellulaires\u00a0\u00bb.   <\/p>\n\n<p class=\"wp-block-paragraph\">Ses travaux portent sur l&rsquo;alpha-synucl\u00e9ine, une prot\u00e9ine intimement associ\u00e9e \u00e0 l&rsquo;apparition de la maladie de Parkinson. Auparavant, Mme Mohamed avait \u00e9tudi\u00e9 la mani\u00e8re dont cet agent s&rsquo;accumule dans le cerveau des souris, mais elle a conclu que les r\u00e9sultats n&rsquo;\u00e9taient pas n\u00e9cessairement applicables \u00e0 ce qui se passe dans le corps humain. Elle s&rsquo;est donc tourn\u00e9e vers les cellules souches extraites du sang de volontaires humains, \u00e0 partir desquelles elle pr\u00e9l\u00e8ve les cellules qu&rsquo;elle utilise pour assembler des mini-cerveaux.  <\/p>\n\n<p class=\"wp-block-paragraph\">\u00ab\u00a0L&rsquo;id\u00e9e est de travailler avec le bagage g\u00e9n\u00e9tique du patient\u00a0\u00bb, explique-t-elle. \u00ab\u00a0Cela permet de personnaliser davantage la pathologie de la maladie. <\/p>\n\n<p class=\"wp-block-paragraph\">Mohamed reconna\u00eet que cette approche, comme l&rsquo;ensemble du domaine des cellules souches, est relativement nouvelle. Elle d\u00e9pend d&rsquo;une combinaison habile de signaux biochimiques et de vaisseaux de stockage sp\u00e9ciaux pour placer ces cellules indiff\u00e9renci\u00e9es \u00e0 proximit\u00e9 les unes des autres afin qu&rsquo;elles deviennent d&rsquo;abord des neurones, puis qu&rsquo;elles les organisent selon des sch\u00e9mas refl\u00e9tant ce que l&rsquo;on pourrait trouver dans un cerveau vivant. <\/p>\n\n<p class=\"wp-block-paragraph\">Elle insiste sur le fait que l&rsquo;effort en vaut la peine, car les mini-cerveaux nous \u00e9clairent d\u00e9j\u00e0 sur la fa\u00e7on dont l&rsquo;alpha-synucl\u00e9ine utilise les cellules c\u00e9r\u00e9brales pour se propager, ce qui constitue la premi\u00e8re \u00e9tape pour comprendre comment la maladie de Parkinson maintient son rythme neurod\u00e9g\u00e9n\u00e9ratif.<\/p>\n\n<p class=\"wp-block-paragraph\">\u00ab\u00a0Ces nouvelles connaissances devraient avoir un impact direct sur le d\u00e9veloppement de nouveaux m\u00e9dicaments destin\u00e9s \u00e0 bloquer la progression de la maladie\u00a0\u00bb, ajoute-t-elle.<\/p>\n\n<p class=\"wp-block-paragraph\">\u00ab\u00a0L&rsquo;id\u00e9e est d&rsquo;\u00e9tudier la propagation de l&rsquo;alpha-synucl\u00e9ine dans un mod\u00e8le de cerveau personnalis\u00e9 et complexe.<\/p>\n\n  D\u00e9couvrez d&rsquo;<a href=\"https:\/\/parkinson.ca\/fr\/category\/profile-expert-fr\/recherche-2018-2020\/\" target=\"_blank\" rel=\"noopener noreferrer\">autres chercheurs<\/a> r\u00e9cemment financ\u00e9s par le programme de recherche de Parkinson Canada en visitant la section recherche du site <a href=\"https:\/\/parkinson.ca\/fr\/recherche\/\" target=\"_blank\" rel=\"noopener noreferrer\">www.parkinson.ca<\/a>.  \n","protected":false},"excerpt":{"rendered":"<p>Nguyen-Vi Mohamed, chercheur postdoctoral \u00e0 l&rsquo;universit\u00e9 McGill, a mis au point des techniques permettant de transformer des cellules souches humaines en neurones, puis de les disposer selon un mod\u00e8le 3D ressemblant au cerveau moyen.
